Symposium 3D STORM au CPN


Au sein de la plateforme NEURIMAG en partenariat avec la société Bruker, un symposium sur l’imagerie Super-résolutive STORM 3D est organisé.

Nous aurons 4 séminaires dédiés au STORM 3D mardi 24 octobre en salle des séminaires (programme ci-dessous).

Le microscope STORM étant présent à l’institut, il est également possible d’avoir une démonstration en petits groupes
ou même de tester vos propres échantillons. Ce microscope permet entre autre l’imagerie à l’échelle de la molécule unique.
Il permet également d’imager les amas de récepteurs/molécules d’adhésion, organelles, vésicules, ou cytosquelette, ... à une résolution de l’ordre de 30 nm.

14h-17h: Workshop on Vutara 3D Single Molecule Localization (SML)with multipoint confocal correlative imaging capabilities. See how the Bruker SRX software Analysis Module turns voxels into data. (Possibility to extend for demonstrations (25/26th of October) with your own samples)

Exploring multiple color 3D STORM microscopy to unravel organels and neuronal architecture

During development and plasticity, synaptic molecules are transported to the synapse via vesicular and endosomal carriers. Fusion of these carriers with the synaptic zone, is achieved by SNARE proteins. We aim to unravel the specificity and contribution of two neuronal v-SNAREs VAMP2 and VAMP7 in the construction and morphogenesis of the synapse at both pre and post-synaptic sites.

Nanoscopy of tripartite synapses

Astrocytes play an active role in shaping and maintaining neuronal circuits through an intimate nanoscale relationship with nearby synapses. We have employed single molecule localisation microscopy to visualise the 3D positions of neurotransmitter receptors and transporters in astrocytes and neurons in fixed cultured cells and brain slices. Moreover, through multi-colour imaging, we assessed the positional relationship between synapses and astroglial receptors and transporters in different conditions compatible with long-term synaptic potentiation or depression.

Combining density-based artifact-free and Bayesian cluster analysis tools for studying the characteristics of membrane-protein organization

The spatiotemporal organization of the membrane proteins are supposed to regulate the cellsignaling activation from outside the cell. Since the proteins are organized on the nanometer-scale, single molecule microscopy as well as very large datasets with hundreds of thousands of localizations are required to understand how the protein organization influences the cell function. We introduce a user-friendly program which comprises a density-based artifact-free approach as well as a Bayesian cluster analysis method in order to study the protein organization. Furthermore, a temporal partitioning for the artifact-free method along with two additional clustering algorithms for the Bayesian cluster analysis have been implemented. In order to solve the computational time and memory problems caused by processing large datasets, we apply a preprocessing step before the Bayesian-based approach. To this end, we suggest splitting the original image into several smaller overlapping regions of interest and stitching them back together after performing the Bayesian cluster analysis.
